Numaferm provides a biochemical production platform for the easy and efficient synthesis of peptides and proteins. This novel technology is called Numaswitch. Target proteins are genetically fused to Switchtag libraries, promoting high-titer production of fusion proteins within E. coli cells as inclusion bodies. Switchtags play a crucial role in facilitating the efficient refolding of fusion proteins, while the unique Numacut TEV protease removes the Switchtag from the targets in a traceless manner. Evolved through artificial intelligence and directed evolution approaches, the Numacut TEV protease demonstrates expanded substrate tolerance compared to the wild type enzyme, allowing the release of targets with native N-termini. Numaswitch addresses the challenge of low refolding efficiencies in inclusion body methods, enabling high product yields and purities. Production of these molecules is accomplished within weeks and in an environmentally sustainable manner, highlighting the technology's advantages.

Numacut TEV Protease

The Numacut TEV protease is an improved variant of the Tobacco Etch Virus (TEV) protease with broadened substrate tolerance in comparison to the wild type enzyme. Evolved by artificial intelligence and directed evolution approaches, the recognition site of the TEV protease (ENLYFQ/S) was broadened to ENLYFQ/X (X: any amino acid, except P). Fusion proteins featuring the ENLYFQ sequence are efficiently cleaved between Q and X releasing N-terminal native target peptides or proteins.

Numaswitch Technology

Numaswitch is a biochemical production platform for easy and efficient peptide and protein production. Target proteins are fused to Switchtags which promote the high-titer production within E. coli cells as inclusion bodies. Switchtags trigger the efficient refolding of fusion proteins, while the Numacut TEV protease removes the desired targets tracelessly from the Switchtag. This technology resolves the challenge of low refolding efficiencies resulting in high product yields and purities.
